Microcephaly is defined as a head circumference of less than 3 standard deviations from the population mean for age and gender. Microcephaly should be considered as a clinical finding rather than a disorder, and it may stem from a variety of causes that lead to the disruption of various stages of brain development. There are both genetic and nongenetic causes of microcephaly.

Canavan disease is a degenerative neurologic condition characterized by aspartoacylase deficiency, resulting in a spongy deterioration of the brain. It is due to mutations in the ASPA gene encoding the aspartoacylase enzyme that catalyzes the conversion of acetylaspartic acid (NAA) to aspartate and acetate. Diagnosis of Canavan disease can be achieved by examining urine NAA.
